Biotinylated mAb cocktail effective for isolating Low Density Granulocytes from SLE patient blood

This University of Michigan group has used a Biotinylated antibody cocktail in conjunction with anti-Biotin MACS beads to quickly isolate and characterize this unusual population of neutrophils from ficoll prepared, RBC-lysed Systemic Lupus Erythematosus patient blood.  Purity of isolated cells was determined as >95% CD10+, CD15+/CD14lo.

 Isolated low density granulocytes (LDG) have capacity to interfere with differentiation of Endothelial precursors.  While LDG have impaired phagocytic function, they are cytotoxic to vascular endothelium. LDG also exhibit enhanced extracellular trap (NET) ability.  This cell population may play a significant role in SLE progression.
